Abstract: Pulmonary arterial hypertension (PAH) is a serious and progressive disease affecting the heart and lungs and is a global health problem affecting individuals and society. In this study, a rat model of PAH induced by monocrotaline (MCT) was successfully constructed. The mitochondrial proteins of each group of rats were determined by using label-free quantitative proteomic techniques. Based on the construction and functional enrichment of protein-protein interaction (PPI) networks, 19 up-regulated mitochondrial genes and 123 down-regulated mitochondrial genes were screened in PAH rats. In addition, in an independent cohort dataset experiment, the results showed that 6 up-regulated mitochondrial genes and 3 down-regulated mitochondrial genes were significantly differentially expressed in the lung tissue of PAH rats. Potential miRNA-targeted central mitochondrial genes were predicted at the transcriptome level using the RNAInter database. The study also identified bortezomib and carfilzomib as potential drugs for the treatment of PAH. Finally, this study sheds new light on key biomarkers and therapeutic strategies for PAHs.

Article Title: PTEN loss confers sensitivity to rapalogs in clear cell renal cell carcinoma

Abstract: Rapalog is an allosteric mTORC1 inhibitor and an approved drug for advanced clear cell renal cell carcinoma (ccRCC). Studies using multiple patient-derived ccRCC cell lines demonstrate that PTEN-deficient cells exhibit hypersensitivity to rapalogs. In PTEN-deficient ccRCC cell lines, Rapalogs inhibited cell proliferation by inducing G0/G1 arrest without inducing apoptosis. Using CRISPR/Cas9-generated isogenic cell lines, the study validated the association between PTEN deletion and Rapallo hypersensitivity. In contrast, deletion of VHL or chromatin-modifying genes (PBRM1, SETD2, BAP1, or KDM5C) did not affect cellular responses to rapalogs. Ectopic expression of an activated mTOR mutant (C1483F) was shown to counteract PTEN-induced cell growth inhibition, while introduction of a drug-resistant mTOR mutant (A2034V) allowed PTEN-deficient ccRCC cells to escape the growth-suppressive effects of rapalogs. PTEN-deficient ccRCC cells were more sensitive to the inhibitory effects of temsirolimus on cell migration and tumor growth in zebrafish and xenograft mice. Studies have shown that PTEN deletion is associated with rapallo sensitivity and can be used as a marker for the selection of rapallo therapy in ccRCC patients.

Article title: Dynamic mechanism of the microbiota of high-salinity organic wastewater with salt-tolerant yeast and its application

Abstract: High salinity organic wastewater is an industrial wastewater with complex components, poor biodegradability and difficult biochemical treatment. In order to reveal the intrinsic mechanism of the microbial community in the treatment of high salinity organic wastewater by salt-tolerant yeast, characterization studies of the microbial community and environmental factors were carried out. The removal rates of chemical oxygen demand, NH3–H and total nitrogen were significantly improved in the pilot-scale WWTP. High-throughput sequencing results showed that biofortification significantly affected microbial community structure. At the phylum level, the Proteobacteria and Ascomycota groups predominate, while at the genus level the Paracoccus and Meyeroi groups predominate. Meyerella plays a key role in the treatment of high salinity organic wastewater, which may be the main reason for the improved pollutant removal efficiency. In addition, pH, dissolved oxygen, and influent volume were key factors affecting the succession of microbial communities during bioaugmentation, however, Meyerrotheca abundance was not affected by influent pollutant concentrations. This study systematically revealed the dynamic changes and functional predictions of microorganisms during bioaugmentation, as well as the relationship between microbial communities and environmental factors, laying a solid theoretical foundation for the subsequent treatment of high-concentration and high-salt wastewater.

Abstract: The reproductive tract of chickens is an important organ for egg formation. The close contact between the vagina and the external environment may lead to the invasion of various pathogenic bacteria, affecting the internal and external quality of eggs, and even increasing the mortality rate and causing economic losses. In recent years, probiotics have brought economic benefits to livestock and poultry production as an alternative to antibiotics. The effects of vaginal administration of B. fragilis on chicken cloacal microbiota, vaginal transcriptome and metabolome were investigated to assess the beneficial potential of B. fragilis. The results showed that B. fragilis treatment affected the microbial composition of the cloaca. Transcriptome analysis found that immune-related genes CCN3, HAS2, and RICTOR were up-regulated, and inflammatory genes EDNRB, TOX, and NKX2-3 were down-regulated, and DEGs were also enriched in the regulation of inflammation, cellular metabolism, and synaptic response pathways. In addition, differential metabolites were mainly related to steroid hormone biosynthesis, unsaturated fatty acid biosynthesis, and arachidonic acid metabolism, and studies identified associations between specific differential metabolites and genes. In conclusion, this study provides a theoretical basis for the application of B. fragilis as a potential probiotic in livestock and poultry production.

Abstract: Cystic echinococcosis (CE) is a neglected tropical zoonotic disease caused by Echinococcus granulosus and remains a major global public health problem. Existing known genotypes of Echinococcus granulosus other than G4 have been identified in China, especially the Qinghai-Tibet Plateau, and their genetic diversity is unique in this region of the world. According to a systematic review, the genetic composition of E. granulosa in my country is as follows: E. granulosa (G1, G3), 98.3%; Echinococcus intermedius (G5), 0.1%; Echinococcus intermedius (G6, G7), 1.4%; and Canada Echinococcus (G8, G10), 0.2%. Specifically, the G1 infection rate was 97.7% and was characterized by a wide host range and geographic distribution. Epidemiological results showed a relatively stable and fine-grained E. coli gene composition in sheep and yak from three CE high-endemic provinces (Xinjiang, Sichuan, and Qinghai). Sheep (287/406, 70.7%) had a higher proportion of fertile cysts than yak (28/184, 15.2%). In the past 29 years, there have been 51 Echinococcus granulosus cox1 haplotypes in China. The ancestral haplotype (Hap_2) remained the most common haplotype, 12 relatively common haplotypes were endemic, and 9 newly reported haplotypes were identified during the survey. The findings suggest that the mandatory immunization of sheep and the EG95 vaccination pilot campaign of yaks closely match the current genotype status.
