Main content: Esophageal squamous cell carcinoma (ESCC) is the main subtype of esophageal cancer. Identifying effective therapeutic targets is of great significance for better treatment of ESCC. Some studies have shown that RNA mismodification may promote the expression and translation of cancer cell-associated mRNAs, but the molecular mechanism remains unclear. This study showed that the tRNA m7G methyltransferase-related complex proteins METTL1 and WDR4 were significantly increased in ESCC tissues and contributed to the poor prognosis of ESCC. Since then, METTL1 and WDR4-related gene knockout studies in ESCC model mice have confirmed that the enhanced activity of tRNA m7G methyltransferase leads to the up-regulation of the expression of these two proteins, which in turn promotes the proliferation of ESCC cells. This study demonstrates an important oncogenic function of misregulated tRNA m7G modification in ESCC and suggests that targeting METTL1 and its downstream signaling pathways may be promising therapeutic targets for ESCC therapy.

With the development of synthetic technology, mRNA has become a new class of therapeutic drugs. However, achieving efficient intracellular delivery of mRNA drugs remains challenging due to the sensitivity of mRNA to RNases. This study proposes a convenient delivery scheme of polyphenol-conjugated nanostructures that can efficiently deliver IL-10 mRNA in vivo. After using this nanostructure to deliver mRNA drugs, IL-10 levels were specifically up-regulated, effectively inhibiting the expression of inflammatory factors, promoting mucosal repair, protecting colonic epithelial cells from apoptosis, and inhibiting dextran sodium sulfate-induced acute and chronic small The murine colitis model has significant therapeutic effects, providing a new gene therapy design for inflammatory bowel disease.

Oxidative stress of sperm is a major factor leading to male infertility, so it is a feasible and effective option to establish male infertility animal models by adjusting the antioxidant system in a targeted manner. However, male germ cell-specific antioxidant mechanisms have not been elucidated. This study confirmed that LanCL1 gene is a specific antioxidant gene in male germ cells, and its expression is reduced or associated with human male infertility. Male mice deficient in the LanCL1 gene exhibit sperm oxidative damage and impaired fertility. In the process of sperm differentiation of germ cells, once the oxidation tendency occurs, transcription factor-specific protein 1 (SP1) will regulate the expression of LanCL1 gene, thereby inhibiting the oxidation process of sperm. This study demonstrates that the SP1-LanCL1 pathway plays a key role in regulating testicular homeostasis and male fertility mediated by redox balance, and suggests that LanCL1-modified mice have attractive applications as an animal model of male infertility.
